![]() It’s easy to assume a man is a man because he is fully grown. So, the idea is that we are easily fooled by appearance. The only line that seems to hint at the core subject matter is the last one in the second verse: “But age doesn’t make you a man.” The lyrics, however, are less forthcoming. The temptation with song, as with others, is to take the title at face value and trust that it is, in fact, a song about young growing old. “They were glued together with body and soul/That much more with their backs up against the wall” is referring to an alliance that stood together until the end. I’m going to take a stab in the dark and say this is a song about war. “I guess it had to happen someday soon/Wasn’t nothing to hold them down/They would rise from among us like a big balloon/Take the sky, forsake the ground.” These lines seem to contradict that notion: Some say this is a song about a couple who endured through hardship. The closing lines, in fact, talk about broken hearts, dreams running dry and the older generation passing on. The hope-filled lyrics of the first verse (“We were ring-around-the-rosy children/They were circles around the sun/Never give up, never slow down/Never grow old, never ever die young”) and second verse do not make a comeback in this song. What’s clear, at least from a surface level, is that everything is hunky dorky in this song until bad things started to happen. There’s more than meets the eye to “Never Die Young” by James Taylor. “Forever Young” by Rod Stewart, A Song About Growing Up (Or Not) That certainly reflects TMBG’s enduring career in some ways too.īut whether you were an “insider” or “outsider” as a youth, maybe you can find yourself in this song. The challenge is in being told that you’re an outsider. While that may be the case, there seems to be a deeper meaning – one where the reference to “youth culture” starts to make more sense.įlansburgh once shared about the idea of insiders and outsiders and said that there’s nothing painful about being an outsider. Flansburgh once said he wrote it as a tribute to his dog Iggy. That is part of their esthetic, however, and I think they like it that way.Īs with all TMBG songs, different comments have been made about “Youth Culture Killed My Dog”. Their songs are impossibly catchy while being quirky and oftentimes difficult to interpret. They Might Be Giants represents the unique and eccentric collaboration between bandmates guitarist John Flansburgh and keyboardist John Linnell. It's okay to maintain a childlike attitude. “Well, when I’m lyin’ in my bed at night/I don’t want to grow up/Nothing ever seems to turn out right/I don’t want to grow up/How do you move in a world of fog/That’s always changing things?/Makes me wish that I could be a dog/Well, when I see the price that you pay/I don’t want to grow up/I don’t ever want to be that way/I don’t want to grow up.” Plainly, it’s a song about how things can get more complex in adulthood. Tom Waits’ “I Don’t Wanna Grow Up” doesn’t have any hidden meanings. While it’s always a good idea to be responsible for yourself, you can still take a childlike (note: not childish) approach to your life. Most people assume that growing up is a necessity and an inevitability. And, if you had parents that cared for you, you probably didn’t have a lot of responsibilities growing up. ![]()

Before the advent of NGS, immune repertoires were largely impenetrable, because it was not possible to enumerate a set of distinct T cells, or clonotypes, large enough to be a meaningful representation of the repertoire in its entirety. A completely analogous process occurs for the TCR α chain, without the D gene segment included.Īs in many areas of investigation, advances in next generation sequencing (NGS) technologies, in which sequences are decoded on arrays and many millions of sequences can be read simultaneously, have been transformative for immune repertoire analysis. The mechanism by which gene segments are joined also introduces base pair variability, which together with the combinatorial selection of these segments results in TCR diversity. Initially one of the two D regions is joined with one of 13 J regions (both randomly selected), followed by joining of the DJ region to one of more than 50 V regions (also randomly selected), yielding a final VDJ region that is approximately 500 bp in length. The TCR-β locus is located on chromosome 7 and is approximately 620 kb in length. (c) Simplified representation of TCR-β VDJ gene recombination resulting in TCR diversity. The CDR3 regions are the main domains of the TCR that are in contact with peptide antigen, and largely determine TCR specificity. Color gradients at junctions represent the regions encoded by arbitrary, untemplated nucleotides introduced during somatic recombination, and which represent a primary source of sequence diversification and TCR variability (see (c) for details). The complementarity determining region 3 (CDR3) domain, approximately 45 nucleotides long, comprises the VJ (for TCR-α) or VDJ (for TCR-β) junction. ![]() In TCR α chains the J region is followed by the V region (orange), whereas in TCR β chains, a D region is located between the V and J regions. T-cell receptor-antigen-peptide-MHC interaction and TCR gene recombination. In this review, we introduce this nascent field and TCR-seq methodology, we discuss recent insights into healthy and diseased TCR repertoires, and we examine the applications and challenges for TCR-seq in the clinic. It is expected that maturation of the field will involve the introduction of improved, standardized tools for data handling, deposition and statistical analysis, as well as the emergence of new and equivalently large-scale technologies for T-cell functional analysis and antigen discovery. However, T-cell repertoire sequencing is still in its infancy. In the context of disease, TCR-seq has been instrumental in characterizing the recovery of the immune repertoire after hematopoietic stem cell transplantation, and the method has been used to develop biomarkers and diagnostics for various infectious and neoplastic diseases. TCR-seq studies have provided new insights into the healthy human T-cell repertoire, such as revised estimates of repertoire size and the understanding that TCR specificities are shared among individuals more frequently than previously anticipated. ![]() High-throughput TCR sequencing (TCR-seq) involves the use of next generation sequencing platforms to generate large numbers of short DNA sequences covering key regions of the TCR coding sequence, which enables quantification of T-cell diversity at unprecedented resolution. GAO was asked to examine the potential cost savings to the government from making changes to currency. Since 1990, GAO had estimated replacing the $1 note with a $1 coin would provide a benefit to the federal government. Other countries have replaced notes with coins of the same value to reduce costs. ![]() Since 2006, both the penny and nickel have cost more to make than their face value. spent about $1.3 billion in 2017 to produce, process, and circulate coins and paper notes for use in the economy. Without such authority, the Mint might not be producing coins as cost-effectively as possible. Since Congress specifies in law which coins are made and their metal composition, the Mint has proposed legislation to enable the Secretary of the Treasury to change the metal content of coins as long as the weight or machine acceptance of the coins is unaffected. Stakeholders were unconcerned about changes to the nickel as long as the changes would not affect how the coin functioned, for example, in vending machines. However, Federal Reserve officials and some stakeholders expressed concern about temporarily suspending the penny due to the potential for external effects, such as penny shortages. ![]() It also estimates that it could save about $74 million over 10 years by changing the metal composition of the dime and quarter. Mint estimates that it could save approximately $250 million over 10 years by suspending penny production and between $2 million and $9 million per year by changing the metal composition of the nickel. For example, armored carriers told GAO that their transportation costs would increase because coins weigh more than notes.Įstimated Cumulative Present-Value Net Loss to the Government from Actively and Gradually Replacing $1 Notes with $1 Coins over 30 Years Seven of 10 stakeholders GAO met with said that replacing the $1 note with a $1 coin would result in additional costs. Stakeholders generally identified few benefits from replacing $1 notes with $1 coins. For example, the lifespan of the $1 note has more than doubled since a 2011 GAO analysis, from 3.3 years to 7.9 years, largely due to changes in note processing technology. GAO's estimates are based on current data and economic projections, which have changed over time. These simulations represent the first time GAO has found that replacing the $1 note with a $1 coin would result in a net loss to the government rather than a net benefit. GAO found the government would incur a loss of about $611 million if notes were actively replaced and about $2.6 billion if $1 notes were replaced gradually (see figure). GAO's analysis found that replacing the $1 note with a $1 coin would likely result in a net loss to the government over 30 years. |
